435EFFECT OF ENALAPRIL ON PHARMACOKINETICS AND PHARMACODYNAMICS OF CHLORPROPAMIDE IN RATSDrug interaction is said to occur when the effects of one drug are changed by the presence of another drug. The main purpose of the study is to evaluate the effect of enalapril on pharmacokinetic and pharmacodynamics of chlorpropamide in rats . Alloxan-induced diabetic model in rats has been used in this study. Diabetes was induced in rats by administering a single dose of intraperitoneal (i. p) injection of alloxan (150mg/kg.), then chlorpropamide and enalapril were administered orally for 7d ays. The pharmacokinetic parameters like t1/2, AUC, clearance, MRT, Tmax and Cmax of chlorpropamide, with and without combination of enalapril treatment were determined. The blood glucose levels were estimated on using Glucose Oxidase - Peroxidase (GOD-POD) method, and serum creatinine levels (Alkaline picrate method) were estimated. Chlorpropamide treatment has shown hypoglycemic and renal impairment activity after oral administration. The increase in chlorpropamide concentration might be due to its interaction with enalapril. Chlorpropamide treatment in combination with enalapril has shown significant decrease in blood glucose levels when compared to rats treated with chlorpropamide alone and chlorpropamide itself shows renal insufficiency, without combination of enalapril.
